Because of the evidence, it appears that Mary Ann is a daughter. I have changed my mind about James and decided he probably is a grandson - after considerable study. The only record linking him to John Mackelroy we have for him is that he witnessed a deed. I believe he is a grandson. It will take me a while to retrace my thoughts as to how I came to this conclusion. However, I have an open mind and will discuss if anyone is interested.
