Legal records show that there were no surviving descendants of Alexander Kincaid, Provost of Edinburgh, nor of his father, Alexander Kincaid, writer in Edinburgh. This is clear because John Kincaid, tenant in Grange was the heir of the Provost's son. With regards to DNA, you are correct in that we then have to go back to John Kincaid, tenant in Grange. Now legal records also subsequently show that the line of the Provost's heir, John Kincaid, tenant in Grange, daughtered out. There remains the possibility that John Kincaid, tenant in Grange, had one or more brothers, but I have yet to identify any yet. All I can clearly say is that there will be no YDNA to be found from legitimate male descendants of Alexander Kincaid, Provost of Edinburgh, nor of his heir, John Kincaid, tenant in Grange. Peter, the way you describe the descendents of Alexander the provost, it looks like the DNA link may not have been broken by his son not having any children himself. If the heir was a grandson of his Alexander the Provost's brother, then any male descendents would share a common ancester and could be identified by DNA matching. So, the question, as always, is how many children did the heir (grandson) have and were any male with further descendents, perhaps some who came to the new world.
